Here is a decent video of Pete Hall at Wordcamp in Portland (2010) demonstrating how to activate the multi-site features in Wordpress.
It get's off to a rough start because of audio difficulties, but it's a good presentation if you are already a Wordpress do it yourself-er and are interested in learning more about using this feature.

It covers minimum requirements and custom functions like domain mapping

Thanks for the link. I'd seen some other videos on posts on setting up multi-site awhile back. This one looks new.
